Seriously, Hoffa and the Mafia each detested the Kennedies each for their own reasons.
Also, Hoffa and the Mafia each used each other for their own benefits.
One particular crime syndicate was assigned to kill Kennedy.
Hoffa had his own dealings with this same family.
He became intimate with all the details during and after. He always kept his mouth shut and kept the union out of it.
While in prison, the Mafia got total control of the Teamsters.
When Hoffa was released from prison, he wanted to return to the Teamsters as president but was thwarted at every turn by the Mafia. They basically threw Hoffa under the bus, because if he returned to power, the Mafia would have lost their control over the Teamsters. It was now an unbelievable cash cow for them. Hoffa's behaviour became erratic and unstable from his rage.
The Teamsters and the Mafia, kept warning him to fade into oblivion quietly. Eventually they gave him an ultimatum when he continued to become more erratic.
He finally threatened to use his Ace up his sleeve, that being he would detail all the facts about the mafia's assasanation of Kennedy.
That's when they made the decision to get rid of Hoffa. Hoffa was too unstable and ready to blow the beans.

The FBI files conclude with all these facts. Officially it's still open but unofficially it's closed in deference to Jimmy Jr.
Jimmy Jr. now president of the Teamsters got in because of the hero-like reputation of his father of the members. The family knows how it happened but the don't want if officially solved because it would expose his father's relationship with the Mafia which would ruin his own credibilty and the cedibilty of the now "clean from corruption" Teamsters. The FBI is also considering it as a courtesy to the family and they also write in their reports that there really is nothing to be gained at this point in time because everyone who they could prosecute is now dead anyway.
In addition, Hoover also had a hatred of the Kennedies as much as Hoffa and the Mafia. The FBI had reports of the impending assasination from when they were keeping tabs on Hoffa, but Hoover, for personal reasons, kept a file but never launched an investigation. He just personally kept abreast of the info he was able to obtain. His enemy's enemy was his fiend in this scenario.
Jack Ruby: Ruby, at the same time, was being considered for a hit by the Mafia for other reasons.. He knew this.
The Mafia told him to kill Oswald or he would be killed. Jail or be killed. Tough choice. He chose jail, obviously.
